In this article the authors created an interaction map of proteins involved in colorectal cancer to look for driver vs. non-driver genes. That is they wanted to see if they could determine what genes are more likely to drive the development and progression in colorectal cancer and which are present in altered states but not necessarily driving disease progression.
The current five-year survival rate of metastasized prostate cancer is only 30% and occurs in every one in nine men. Researchers have shown that people with a type of dwarfism called Laron’s Syndrome are immune to cancer due to their low levels of insulin-like growth factor-1 (IGF-1). For this reason, experimentally modifying the level of IGF-1 could provide better insight into whether lowering the levels of IGF-1 in prostate cancer cell lines (e.g. PC-3) could be an effective treatment to reduce their rates of proliferation and migration and increase apoptosis. We selected three compounds, which researchers have shown decrease IGF-1 levels, to test and combine to determine which is the most promising.
Osteosarcoma is a type of bone cancer that affects young adults and children. Early diagnosis of osteosarcoma is crucial to successful treatment. The current methods of diagnosis, which include imaging tests and biopsy, are time consuming and prone to human error. Hence, we used deep learning to extract patterns and detect osteosarcoma from histological images. We hypothesized that the combination of two different technologies (transfer learning and data augmentation) would improve the efficacy of osteosarcoma detection in histological images. The dataset used for the study consisted of histological images for osteosarcoma and was quite imbalanced as it contained very few images with tumors. Since transfer learning uses existing knowledge for the purpose of classification and detection, we hypothesized it would be proficient on such an imbalanced dataset. To further improve our learning, we used data augmentation to include variations in the dataset. We further evaluated the efficacy of different convolutional neural network models on this task. We obtained an accuracy of 91.18% using the transfer learning model MobileNetV2 as the base model with various geometric transformations, outperforming the state-of-the-art convolutional neural network based approach.
In this article, the authors analyzed ribosome profiling data from amino acid-starved pancreatic cancer cells to explore whether the pattern of ribosome distribution along transcripts under normal conditions can predict the degree of ribosome stalling under stress. The authors found that ribosomes in amino acid-deprived cells stalled more along elongation-limited transcripts. By contrast, they observed no relationship between read density near start and stop and disparities between mRNA sequencing reads and ribosome profiling reads. This research identifies an important relationship between read distribution and propensity for ribosomes to stall, although more work is needed to fully understand the patterns of ribosome distribution along transcripts in ribosome profiling data.
Smoking generates free radicals and reactive oxygen species which induce cell damage and lipid peroxidation. This is linked to the development of oral cancer in chronic smokers. The authors of this study developed Quitpuff, simple colorimetric test to measure the extent of lipid peroxidation in saliva samples. This test detected salivary lipid peroxidation with 96% accuracy in test subjects and could serve as an inexpensive, non-invasive test for smokers to measure degree of salivary lipid peroxidation and potential risk of oral cancer.
Acquired drug resistance is an increasing challenge in treating cancer with chemotherapy. One mechanism
behind this resistance is the increased inflammation that supports the progression and development of
cancer that arises because of the drug’s presence. Integrative oncology is the field that focuses on including natural products alongside traditional therapy to create a treatment that focuses on holistic patient well-being.
In this study, the authors demonstrate that the use of an herbal formulation, consisting of turmeric and green tea, alongside a traditional chemotherapeutic drug, 5-fluorouracil (FU) significantly decreases the level of cytokines produced in breast cancer cells when compared to the levels produced when exposed solely to the chemo drug. The authors conclude that this combination of treatment, based on the principle of integrative oncology, shows potential for reducing the resistance against treatment conferred through increased inflammation. Consequently, this suggests a prospective way forward in improving the efficacy of cancer treatment.
This study follows the process of single-cloning and the growth of a homogeneous cell population in a superficial environment over the course of six weeks with the end goal of showing which of five tumor growth models commonly used to predict heterogeneous cancer cell population growth (Exponential, Logistic, Gompertz, Linear, and Bertalanffy) would also best exemplify that of homogeneous cell populations.
Sesame (Sesamum indicum) and moringa (Moringa oleifera) have natural antioxidants that could prevent cancer growth. Previously, this group found that sesame and moringa individually suppress eye tumor grown in the Drosophila melanogaster model. In the present study, combinations of sesame and moringa at different concentrations were included in the D. melanogaster diet. The impact on eye tumor development was assessed at different stages of growth.
The authors investigate whether human blood cancers carrying mutations in DNA repair genes possess increased sensitivity to common chemotherapy drugs cisplatin or gemcitabine.
The authors use HF-1, an herbal formation, on bone marrow derived cells as well as breast cancer cells to assess HF-1's ability to prevent tumorigenesis. As metastasis requires coordination of multiple cells in the tumor microenvironment, their findings that HF-1 augments cytokine expression such as VEGF & TGF-B show that HF-1 has potential application to therapeutics.
