Wound healing properties of mesenchymal conditioned media: Analysis of PDGF, VEGF and IL-8 concentrations

Regenerative medicine has become a mainstay in recent times, and employing stem cells to treat several degenerative, inflammatory conditions has resulted in very promising outcomes. These forms of cell-based therapies are novel approaches to existing treatment modalities. In this study, the authors compared the concentrations of the cytokines PDGF, IL-8, and VEGF between conditioned and spent media of mesenchymal stem cells (MSCs) to evaluate their potential therapeutic properties for wound healing in inflammatory conditions. They hypothesized that conditioned media contains higher concentrations of wound healing cytokines compared to spent media. The authors found that while IL-8 and VEGF were present in highest concentrations in conditioned media, PDGF was present in maximal amounts in spent media.

Here, recognizing that the immune response to cancer results in biomarkers that can be used to assess the immune status of cancer patients, the authors investigated the concentrations of key cytokines (TH1 and TH2 cytokines) in healthy controls and cancer patients. They identified significant changes in resting and activated cytokine profiles, suggesting that data of biomarkers such as these could serve as a starting point for further treatment with regard to a patient's specific immune profile.

Estimation of cytokines in PHA-activated mononuclear cells isolated from human peripheral and cord blood

In this study, the authors investigated the time-dependent cytokine secretion ability of phyto-hemagglutinin (PHA)-activated T cells derived from human peripheral (PB) and cord blood (CB). They hypothesized that the anti-inflammatory cytokine, IL-10, and pro-inflammatory cytokine, TNFα, levels would be higher in PHA-activated T cells obtained from PB as compared to the levels obtained from CB and would decrease over time. Upon PHA-activation, the IL-10 levels were relatively high while the TNFα levels decreased, making these findings applicable in therapeutic treatments e.g., rheumatoid arthritis, psoriasis, and organ transplantation.

This study focused on finding more sustainable growing methods that reduce chemical fertilizer or water usage and can be used at the household level for garden plants. Metrics for healthy plant growth were height at first bloom, growing time, and survival rate. The Deep Water Culture (DWC) treatment for garden cress plants significantly increased the height at first bloom compared to the control group. For rates of surviving plants, the treatments had little effect on garden cress, but the Eggshell Grounds, Wick System, and DWC system groups outperformed the control group for marigolds.

This study tested the proficiency of different concentrations of the antimicrobial sorbic acid to inhibit the probiotic Lactobacillus casei Shirota. It was hypothesized that sorbic acid’s use as a bacterial deterrent would also target this bacterial strain of Lactobacillus. The results supported the hypothesis, with the colony count of L. casei Shirota having significant decreases at all concentrations of sorbic acid. These results additionally suggest that even under the FDA sorbic acid restrictions of 0.03% concentration, damaging effects could be seen in L. casei Shirota.

Application of gene therapy for reversing T-cell dysfunction in cancer

Since cancer cells inhibit T-cell activity, the authors investigated a method to reverse T-cell disfunction with gene therapy, so that the T-cells would become effective once again in fighting cancer cells. They used the inhibition of proprotein convertases (PCSK1) in T cells and programmed death-ligand 1 (CD274) in cancer cells. They observed the recovery of IL-2 expression in Jurkat cells, with increased recovery noted in a co-culture sample. This study suggests a novel strategy to reactivate T cells.

With climate change and rising sea levels, south Brooklyn is exposed to massive flooding and intense precipitation. Previous research discovered that flooding shifts plant species distribution, decreases soil pH, and increases salt concentration, nitrogen, phosphorus, and potassium levels. The authors predicted a decreasing trend from Zone 1 to 6: high-pH, high-salt, and high-nutrients in more flood-prone areas to low-pH, low-salt, and low-nutrient in less flood-prone regions. They performed DNA barcoding to identify plant species inhabiting flood zones with expectations of decreasing salt tolerance and moisture uptake by plants' soil from Zones 1-6. Furthermore, they predicted an increase in invasive species, ultimately resulting in a decrease in biodiversity. After barcoding, they researched existing information regarding invasiveness, ideal soil, pH tolerance, and salt tolerance. They performed soil analyses to identify pH, nitrogen (N), phosphorus (P), and potassium (K) levels. For N and P levels, we discovered a general decreasing trend from Zone 1 to 6 with low and moderate statistical significance respectively. Previous studies found that soil moisture can increase N and P uptake, helping plants adopt efficient resource-use strategies and reduce water stress from flooding. Although characteristics of plants were distributed throughout all zones, demonstrating overall diversity, the soil analyses hinted at the possibility of a rising trend of plants adapting to the increase in flooding. Future expansive research is needed to comprehensively map these trends. Ultimately, investigating trends between flood zones and the prevalence of different species will assist in guiding solutions to weathering climate change and protecting biodiversity in Brooklyn.

Asthma affects over 334 million people worldwide and is triggered by inhalation of environmental stimuli. The authors of this study characterized the effect of exposure to common spoilage yeast, Pichia kidriavzevii on alveolar epithelial cells. A direct correlation between infection duration and asthmatic status of these cells was found, indicating the potential for this yeast to be an environmental stimulus of asthma and warranting further study.
